I'd say they're both pretty great, consistently hilarious movies that are also impressively sharp and smart in their American satire, and this is coming from someone who doesn't think much of South Park the show (The Simpsons is/was of course great). Overall I'd probably give the edge to The Simpsons Movie, or though I'm not sure if that's because it's a stronger film or if it's just more goodwill carried over from the show.
Either way, they're both significantly better than they probably had any right to be.

I gotta give the edge to Trey Parker/Matt Stone here. They really knocked it out of the park and pushed the envelope for what they could get away with.

South Park: Bigger, Longer, and Uncut is funnier and I watched it more often when I was a kid, but nowadays I think I'd probably have more interest in The Simpsons Movie if only because it felt like they really captured some of the magic and heart of the show's golden age.

The South Park one is pure majesty. The Simpsons one was very good too, but it doesn't have a patch on Bigger, Longer & Uncut.
