Two very talented guys who broke a lot of ground ......I know Murphy will most likely win because maybe people don't go back as far with Richard Pryor and he ended badly but damn I would love to see Murphy in something like Blue Collar.

What I've seen from Pryor is great comedy skills. Maybe I haven't seen enough of him, I must admit, but Murphy can do more than just comedy.
Not really talking about his action flicks, his acting chops in drama are quite special.

Stand-up: Pryor, even though Murphy was phenomenal. Pryor's simply the GOAT standup.
Acting: Murphy

Acting: Murphy I voted for Pryor just because he is the GOAT at stand-up, and because he inspired so many comedians after him. For acting probably Murphy just because as TerryMontana mentioned Murphy has succeeded in more than comedies, but also because I've seen way more of Murphy's films than I have of Pryor's. I haven't really sought out Pryor's films, but I remember watching a lot of his movies with my dad when I was younger like Brewster's Millions, The Toy, and most of the movies that he made with Gene Wilder. Beyond those, I also remember him in a few other movies like Superman III, The Wiz, Lost Highway, etc., but either I didn't like those films or he didn't really stand out, or didn't stand out in a good way, that they didn't immediately come to mind when I remember him.

Pryor also never had his own "great comedy film" like Eddie Murphy did with Beverly Hills Cop, and Coming to America, that allowed to really showcase his strengths as a performer. His best films were when he was paired / playing second fiddle to Gene Wilder ( Silver Streak, Stir Crazy), or his rare moments of drama ( Blue Collar, which he is fantastic in). Sure his films made money, but I would argue a lot of movies misused Pryor in the 80s, especially when he was trying to go for a more "family friendly image", in the wake of his fire-accident, like The Toy. I have a soft spot for Brewster's Millions but I acknowledge it's not top comedy. There's a reason his concert films tend to be his best regarded work.

Richard Pryor is the greatest. Eddie was great too, but no one beats Pryor to me.

Pryor was a better stand-up. But Murphy isn't that far behind him
As a film actor and film comedian, Murphy is leagues ahead of Pryor. It really isn't close. So much of Pryor's comedic film work dates badly, whereas the best of Murphy like Trading Places and Beverly Hills Cop are just as funny today as when they were released, mainly because Murphy was such a remarkable film comedic actor. He has a tonne of range.
You can watch Pryor bumbling around being unfunny in Superman 3 today, and genuinely wonder why anyone considered him a comedic genius. His stand-up showcased him way better than his movies.
Even in dramatic parts, Murphy has more ability.
So overall, Murphy easily.

I never watched either guys stand-up in any detail, but I love them both just for their movie work. Murphy in his prime was something special. I mean he was a Robin Williams like force of nature on screen, and he's got charisma for days. Even when middle aged, rich, has been Eddie Murphy came out briefly on that SNL 40th episode the other year he was still the coolest fucker in the room without even doing anything.
Still, Pryor for me could just conjure up so much more vulnerability on screen. Yes, a lot of the movies sucked, but he was always so god damn endearing.

Yeah, this is a good summary, though I would argue that some of Pryor's comedic stuff is less dated, and more that he was often working with crummy scripts. Murphy had this same problem later on in his career, but he at least came running strong out of the gate with 48 Hrs and Trading Places. I'd say that Pryor's best years were in the seventies. Things like The Mack, and Bingo Long and The Traveling All-Stars, films where he's not necessarily the focus of the film, but he is allowed to be funny comic relief, and the movies themselves are actually pretty solid too (if you haven't seen The Mack, I recommend it). When he became a leading man after Silver Streak, it seems like he fell into that trap that every other comedians do, in that the studios would often handed him half-baked screenplays with ludicrous premises, that they expected him to elevate with ad-libbing, and comedic charisma (the atrocious comedy known as Critical Condition is a great example of this). Predictably 90% of the time the results would be bad. An interesting parallel / comparison between Murphy and Pryor is that both The Toy, and 48 Hrs. opened on the same day. The Toy actually out-grossed Hrs on opening weekend, but the later was longer-lasting in theaters and made more.

Pryor couldn’t pull off the multiple-character films Eddie did in Coming to America and The Nutty Professor nearly as effective as Eddie could. Pryor’s domain was stand-up and he was GOAT level at it. Everything else (tv sketch comedy, movies)..Murphy by light years.
