Not just movies, not just TV, not just stand-up comedy, not just radio DJ's, not just newspaper cartoonists, not just talk show hosts or talk show guests...we're including ALL OF IT in 1 giant tent and listing the ten people who you think have most made you laugh throughout the years (or, if you prefer, the ten funniest people ever).
Ranking this list is optional, but I'm gonna go ahead and list mine in alphabetical order. It just so happens the first one listed in that order would definitely be my #1 choice for funniest person ever, but the rest would be to hard to rank by preference.

Can't settle on a definitive top ten, but my number one is probably John Swartzwelder. Phil Hartman would probably be second.
Quick edit: Norm Macdonald is way up there too.

Jennifer Saunders is up there in those classic sketches of hers. In recent times, Kudrow and Coolidge are totally disarming.
The funniest actor I've ever seen was Alejandro Urdapilleta, underground Argentine actor. That guy came from a different solar system.
