You probably already know that she was director Mike Nichols' first choice for the role. He visited her at her home in London to discuss the film, but ultimately she turned him down. I love Anne Bancroft's performance (it's iconic for a reason), but I do think Gardner would have been a more age-appropriate choice. Bancroft was only 6 years older than Hoffman... And around this time, Gardner really seemed to have come into her own as an actor. T he Night of the Iguana is by far her best, most relaxed performance, don't you think? Can you imagine her here? Or is Bancroft's performance just too special to even think of anyone else?

That's actually the opposite of what I've always heard From IMDB and Wikipedia I've heard both Doris Day and Jeanne Moreau cited as the first choice for Mrs. R. As for alternate castings, it's interesting to think about them going sort of the opposite direction and casting a sophisticated European in the role, someone like the aforementioned Moreau, Audrey Hepburn (who lobbied for the role), Deborah Kerr, or Claire Bloom. But ultimately Bancroft is too good to actually want someone different in the role. The movie is perfect as is.

I'm trying to picture Doris Day in the part after reading Countjohn's post. It's not too easy to imagine, but I do think she would have been a very sensual Mrs. Robinson.
On Ava, I think she would have been good in the role. Of course, Anne Bancroft is such a class actress that I wouldn't want to replace her, but I think Gardner could have done really well as Mrs. Robinson. That would have been great for her careerwise too to have an iconic role like that in the late 60s.
