Watching Bull Durham (surprise, surprise) and I love how great Costner and Sarandon play off one another and yet they never worked again (if I am wrong here please correct me).
Then there's a supreme odd couple of Cage and Connery in The Rock. Two Oscar winners in a Michael Bay movie who miraculously WORK. Maybe it's hindsight bias, but this has to be The Odd Couple of odd couples.

First that come to mind are Cary Grant and Jimmy Stewart. Two of my all time favs who made a great duo in Philadelphia Story and never worked together after that.

Cage & Hunter - Raising Arizona Pacino has a ton of terrific "1 time chemistry" ones (in some cases "1 scene") with Oscar winners especially too (Streep, DiCaprio, Mirren, Irons, Lemmon, Walken, Hackman, Pesci, Brando, Holly Hunter, Robin Williams, Penn, etc.) but "surprisingly" to me would be with Russell Crowe. I'm not the biggest Crowe fan - though I really like his work from ~91-01 - and I don't find him particularly special at interacting with others.......but I love them together in The Insider (1999) and would have thought those two would clash in a very bad way.

Denzel and Sarita Choudhury in Mississippi Masala. You could make a solid argument that Sarita is Denzel’s best love interest in film. Their on screen chemistry is amazing.
Would’ve said Julia Roberts a few weeks ago but since their new project together got announced can’t use her anymore.

They were even great friends in real life so there was no off camera reason why. Would have much rather seen her with Peck than some of her later leading men. Bogart and Bergman is another obvious one for a classic romantic couple. Although they were at each other's throats the whole Casablanca shoot so that was probably why they never did another movie. First that come to mind are Cary Grant and Jimmy Stewart. Two of my all time favs who made a great duo in Philadelphia Story and never worked together after that. That's one I didn't think of on my own because I guess I sort of assumed they did another one at some point. They were friends in real life and there are tons of quotes of them praising each other in interviews, plus P. Story is a signature role for both of them, so you would have thought they'd have paired another time. Similar situation with Stewart and Katharine Hepburn who I don't think worked together after the movie either.

There have been a lot of speculation about that era in Hollywood, particularly concerning Audrey Hepburn and Grace Kelly, that they had more than a few affairs on-set with their leading men (many of whom were married). Some confirmed, some rumoured. They always denied it, but Peck and Hepburn were rumored to have hooked up. Since Peck was married, a likely explaination why they never worked together again is that his wife at least caught wind of the rumours and nixed the idea of her husband working with Hepburn again (who did have a confirmed affair with William Holden on Sabrina)

Indeed K.Hep and Stewart never worked together again although they had great chemistry in PS. If I'm not mistaken, Hepburn worked only once with many big stars back then (Bogart, Olivier, O'Toole). On the other hand she made two movies with Grant.
