Matthew McConaughey - Magic Mike. Whether you want to count it as a breakthrough or not, but it set the stage that would bring us Dallas Buyers Club, Mud, TRUE DETECTIVE, Interstellar, etc.

Gonna have to do a top 10, both genders:
1) Rosamund Pike, Gone Girl (could argue she was already on the map, but this shot her to another level) 2) Timothee Chalamet, Call Me by Your Name 3) Vicky Krieps, Phantom Thread 4) Jennifer Lawrence, Winter's Bone 5) Trevante Rhodes, Moonlight 6) Alicia Vikander, Ex Machina 7) Rooney Mara, The Girl with the Dragon Tattoo 8) Florence Pugh, Lady Macbeth 9) Daniel Kaluuya, Get Out 10) Brie Larson, Short Term 12
HM: Emma Stone, Easy A Hailee Steinfeld, True Grit Ezra Miller, We Need to Talk About Kevin Aisling Franciosi, The Nightingale

Elizabeth Olsen in Martha Marcy May Marlene is my top choice. I loved her performance and also the ending to that film.

2. Saoirse Ronan- Brooklyn her breakthrough was in Atonement or hanna There are different degrees of breakthroughs. It was really her first successful adult part and made that transition for her after kind of a gap in her career in the first half of the 2010's.
